You'll have a blast just wandering around, but I'd start out in Chelsea, work your way down to the Meat Packing District (take the HIGHLINE if it's nice weather--you can ask anyone where it is), then over to the West Village, and then you can either go south for great shopping in Soho, or keep moving east for awesome little restaurants and shops in the East Village. I definitely suggest wandering with a map--you'll find the structure of the city isn't hard to figure out once you're out and about, and the subways make it super easy to get around. I lived in San Juan for a year, back when I was in my 20s. I never ran into any anti-gay sentiment in the city. I think if you go to other Puerto Rican cities, you might not find any gay presence at all. Also, you may not hear any English. But you can do day trips to most of the outlying sightseeing locations (such as El Yunque Rain Forest or Rio Camuy Caves) and still be back in San Juan before the end of the day. The weather is usually dependably in the mid-80s during January and February.
